What is in a Semax nasal spray?

The peptide is the same one we sell as a 10 mg lyophilised vial: the ACTH(4-7) fragment Met-Glu-His-Phe joined to a Pro-Gly-Pro tail that resists peptidases, developed at the Institute of Molecular Genetics of the Russian Academy of Sciences. The difference is the form. The vial is dry powder; the spray bottle holds 20 mg of Semax dissolved in 20 mL, a nominal 1 mg per mL, so no reconstitution step is involved.

The nasal route is the one the Semax literature mostly uses. The developers’ own review describes intranasal administration in animals and people, and a 2006 kinetics study with tritium-labelled Semax detected labelled material in rat brain two minutes after intranasal application, most of it still intact Semax. That is a feature of the research record, not a recommendation: this bottle is a laboratory reference material.

Semax identity

Names Semax, ACTH(4-7)-Pro-Gly-Pro, ACTH(4-7)PGP; Semax nasal spray
Sequence MEHFPGP (H-Met-Glu-His-Phe-Pro-Gly-Pro-OH)
Molecular formula C37H51N9O10S
Molecular weight 813.9 g/mol
CAS number 80714-61-0
PubChem CID 9811102
This product Solution, 20 mg Semax in 20 mL (nominal 1 mg/mL), spray bottle

This is unmodified Semax. “N-acetyl Semax amidate” is a different molecule (Ac-MEHFPGP-NH2); see the Semax vial page for the difference.

Certificate for the current lot

Every bottle is filled from one lot and ships against that lot’s certificate, issued by HYTE Research Division. Identity is confirmed by HPLC with diode-array detection, and the Semax concentration is measured against a specification of not less than 90% of the label claim:

The spray certificate reports identity and concentration; it does not report an HPLC purity figure. The 10 mg vial has its own certificate, from ILS Laboratories, with an HPLC purity result.

How Semax has been studied

The clinical Semax literature is Russian, mostly in stroke rehabilitation and mostly in Russian. ClinicalTrials.gov lists no Semax study.

Regulatory status

A 2022 review lists Semax among the peptide products registered in the Russian Federation, as a nootropic. In the US it is not an approved drug; FDA’s compounding safety-risk page lists “Semax (heptapeptide)” among nominated-but-withdrawn substances. Deigin VI, et al. Pharmaceutics, 2022; FDA compounding safety-risk list. Clean Peptide’s Semax Spray is for laboratory research only, not for human or veterinary use.

QuestionsSemax Spray: frequently asked

What is in the Semax nasal spray?+

20 mg of Semax (MEHFPGP) dissolved in 20 mL, a nominal 1 mg per mL, in a spray bottle. The current lot’s certificate confirms identity by HPLC-DAD and reports the measured concentration.

What is the difference between Semax Spray and the Semax vial?+

Same peptide, different form. The vial is 10 mg of lyophilised powder with an ILS Laboratories certificate that includes HPLC purity; the spray is 20 mg already in solution with a HYTE Research Division certificate for identity and concentration.

Was Semax studied as a nasal spray?+

The published Semax research mostly used intranasal application, in rats and in Russian clinical studies, and a 2006 study tracked labelled Semax into rat brain after intranasal application.

Is this N-acetyl Semax amidate?+

No. This is unmodified Semax, C37H51N9O10S (813.9 g/mol, CAS 80714-61-0). N-acetyl Semax amidate is a separate molecule with an acetylated N-terminus and an amidated C-terminus.

Is Semax FDA-approved?+

No. A 2022 review lists it as registered in Russia as a nootropic. FDA lists Semax among bulk substances nominated, then withdrawn, from its compounding safety-risk category.
