DSIP Spray Delta sleep-inducing peptide
DSIP (delta sleep-inducing peptide, emideltide) is the nonapeptide Trp-Ala-Gly-Gly-Asp-Ala-Ser-Gly-Glu, first isolated from rabbit blood in 1977. This DSIP nasal spray holds 20 mg in a 20 mL bottle, already in solution, with a lot certificate.

What is DSIP?
Delta sleep-inducing peptide was isolated in Basel by Schoenenberger, Monnier and colleagues from the cerebral venous blood of rabbits whose thalamus was being electrically stimulated to induce sleep. Infused into the brains of other rabbits, the peptide increased slow-wave (delta) EEG activity, which gave it its name. Its sequence, WAGGDASGE, was reported in 1977 and synthesised in 1978.
Almost fifty years on, DSIP is still something of a puzzle. No gene, precursor protein or receptor for it has been isolated, and a 2006 review in the Journal of Neurochemistry called it “a still unresolved riddle”. Its structure does not resemble any other known peptide family.

Published research on DSIP
- Discovery (rabbits, 1977). The isolated peptide enhanced delta and spindle EEG activity under double-blind conditions; of nine synthetic peptides tested, only synthetic DSIP did so. Characterization of a delta-electroencephalogram (-sleep)-inducing peptide. Schoenenberger GA, Monnier M. PNAS, 1977.
- Sequence and synthesis (1978). Synthetic DSIP raised delta activity by a mean of 35% in neocortex and limbic cortex; only the alpha-aspartyl form was highly active, not its beta-Asp isomer. The delta EEG (sleep)-inducing peptide (DSIP). XI. Amino-acid analysis, sequence, synthesis and activity of the nonapeptide. Schoenenberger GA, et al. Pflügers Archiv, 1978.
- Crossing the blood-brain barrier (rabbits, 1977). Given intravenously, synthetic DSIP increased cortical delta activity and reduced motor activity over five hours. Transport of the synthetic peptide DSIP through the blood-brain barrier in rabbit. Monnier M, et al. Experientia, 1977.
- Review of the first decade (1984). DSIP-like material was detected in brain, peripheral organs and plasma, with reported effects on EEG, neurotransmitters, circadian patterns and hormone levels and a U-shaped dose-response. Delta-sleep-inducing peptide (DSIP): a review. Graf MV, Kastin AJ. Neuroscience and Biobehavioral Reviews, 1984.
- The human studies disagree. Small intravenous studies in the 1980s reported sleep effects in people with insomnia; a 1992 double-blind study in 16 patients found only weak effects and concluded short-term DSIP was unlikely to be of major benefit. Schneider-Helmert D, et al. Neuropsychobiology, 1983; Effects of delta sleep-inducing peptide on sleep of chronic insomniac patients. A double-blind study. Bes F, et al. Neuropsychobiology, 1992.
- Where the field stands (2006). The link between DSIP and sleep was never characterised further, partly because no DSIP gene, protein or receptor has been found; the authors propose that an unidentified DSIP-like peptide underlies the activity. Delta sleep-inducing peptide (DSIP): a still unresolved riddle. Kovalzon VM, Strekalova TV. Journal of Neurochemistry, 2006.
The published DSIP studies used infusion into the brain or intravenous injection. We did not find a published study of nasal administration.
Regulatory status
DSIP is not an approved drug in the US. FDA’s compounding safety-risk page lists “Emideltide (DSIP)” among nominated-but-withdrawn substances. A 2022 review lists a Russian preparation with the DSIP sequence, Deltaran, among peptide products registered there. FDA compounding safety-risk list; Deigin VI, et al. Pharmaceutics, 2022. Clean Peptide’s DSIP spray is for laboratory research only.

QuestionsDSIP Spray: frequently asked
What is DSIP?+
DSIP, delta sleep-inducing peptide (emideltide), is the nonapeptide Trp-Ala-Gly-Gly-Asp-Ala-Ser-Gly-Glu, isolated from rabbit blood by Schoenenberger and Monnier in 1977.
What is in the DSIP nasal spray bottle?+
20 mg of DSIP in 20 mL of solution. The current lot’s certificate confirms identity by HPLC-DAD and reports the measured concentration.
Does DSIP have a known receptor?+
No. No DSIP gene, precursor protein or receptor has been isolated, which is why a 2006 review called it an unresolved riddle.
What is the DSIP molecular weight?+
848.8 g/mol (C35H48N10O15, PubChem CID 68816). PubChem lists CAS 62568-57-4.
Was DSIP studied as a nasal spray?+
Not that we could find. The published studies gave DSIP by infusion into the brain or by intravenous injection.
