What is retatrutide?

Retatrutide is a 39-residue synthetic peptide developed by Eli Lilly from the GIP backbone. A single molecule activates three receptors: the glucose-dependent insulinotropic polypeptide receptor (GIPR), the glucagon-like peptide-1 receptor (GLP1R) and the glucagon receptor (GCGR). In Lilly’s discovery paper it showed balanced glucagon- and GLP-1-receptor activity with more GIP-receptor activity in vitro. It carries a C20 fatty diacid on lysine 17, which binds albumin and gives it a long half-life: about 6 days in a phase 1b study.

It is the third molecule in a progression from Lilly and Novo Nordisk: semaglutide acts on one receptor (GLP-1R), tirzepatide on two (GIPR and GLP-1R), and retatrutide on three. See retatrutide vs tirzepatide and retatrutide vs semaglutide.

Why “GLP-3 Reta”?

“GLP-3” is a shop label, not a scientific name. There is no GLP-3 hormone or receptor; Lilly itself calls the term a scientifically inaccurate label used informally for triple agonists. Research catalogues adopted “GLP-3 R” and “GLP-3 RT” for retatrutide because it adds a third receptor to the GLP-1 class. Retatrutide is an investigational drug. It has not been approved by FDA or any other regulator; press coverage of Lilly’s July 2026 announcement reports that Lilly plans to submit it to FDA in 2027.
